WAY 207024 dihydrochloride是一种有效的促性腺激素释放激素受体(GnRH-R)拮抗剂,作用于人和大鼠的GnRH受体,IC50值分别为12和71 nM[1]。
GnRH-R是G-蛋白偶联受体(GPCR)家族的成员,通过结合促性腺激素释放激素而被激活,引起垂体前叶释放黄体生成素(LH)和卵泡刺激素(FSH),它们反过来刺激性激素的合成和释放进入全身循环[1]。
WAY 207024 dihydrochloride具有中度的膜渗透性和大鼠肝微粒体稳定性[1]。
在阉割大鼠中,WAY 207024 dihydrochloride口服给药后吸收良好,有74%的生物利用度。WAY 207024 dihydrochloride(30 mg/kg,口服灌胃)在给药后的1小时开始减少血清LH的水平,而在24小时后回复到对照组水平[1]。
