TRAM-39,2-(2-氯苯基)-2,2-二苯基乙腈,是一种有效的和合成的中间电导Ca2+激活K+通道选择性抑制剂。TRAM-39可选择性阻断K-Ca3,但不能抑制其他Ca2+活化的K+通道。[1,3]
中介电导Ca2活化的K通道是IKCa1基因的产物,对调控结肠上皮细胞和淋巴细胞膜电位非常重要。IKCa1基因对红细胞的体积调节也十分重要。IKCa1可调控paneth细胞alpha-防御素释放,参与肠上皮细胞对摄入的细菌病原体的黏膜防御。[1,2]
TRAM-39可抑制Kca3,以抑制细菌或细菌脂多糖刺激的paneth细胞分泌。内皮衍生的超极化因子(EDHF)响应可通过活化内皮SKCa和IKCa引发。[1,3]
TRAM-39和apamin的结合可消除内皮衍生的超极化因子(EDHF)介导的大鼠来源的内皮细胞松弛。TRAM-39和apamin的结合可诱发微弱和短暂的内皮细胞去极化。[2]
