SU5614是一种蛋白酪氨酸激酶抑制剂。
酪氨酸激酶是通过信号转导级联活化多种蛋白的酶。通过向蛋白中加入磷酸基团来激活蛋白。
体外:之前的研究发现,SU5614可诱导表达c-kit的Kasumi-1、M-07e和UT-7细胞的生长停滞和细胞凋亡,并抑制干细胞因子(SCF)诱导的对c-kit的酪氨酸磷酸化。此外,Kasumi-1细胞对SU5614生长抑制活性的敏感性主要是由SCF的自分泌产生引起的,而不是由c-kit的转化突变引起的[1]。
体内:小鼠给药SU5614(一种血管内皮生长因子(VEGF)抑制剂),甲苯二异氰酸酯吸入72小时后在BAL液中VEGF的水平显著降低。此外,与酶免疫实验获得的结果一致,蛋白印迹分析显示,在甲苯二异氰酸酯吸入72小时后,SU5614降低BAL液中VEGF的水平。这些结果表明,VEGF可能是甲苯二异氰酸酯诱导哮喘的主要决定因素之一,抑制VEGF可能是一个很好的治疗策略[2]。
临床试验:到目前为止,SU5614仍处于临床前研究阶段。
