Recommended Dilution:
1:20 to 1:50 dilution with PBS containing 10% normal goat serum. Working dilution for specific application should be determined by individual investigator to obtain the best conditions.
1)Q:Streptavidin-Cy3、Streptavidin-Cy5、Streptavidin-FITC三种产品应该如何选择?
A:三种产品的不同点在于荧光颜色不同,您可根据实验需要来进行选择,应尽量减少荧光染料间的光谱重叠。FITC是广泛使用的荧光基团,但其最大的缺点是淬灭快,使用抗淬灭剂可以减轻;Cy3和Cy5比大多数荧光基团更亮,更稳定,背景更弱,二者常在共聚焦显微实验中作多标记;Cy5的缺点是不能用表面荧光显微镜观察,需用具有远红外检测器的共聚焦显微镜观察。
2)Q:免疫荧光实验时背景过高?
A:可能原因及解决方案如下:
(1) 一抗浓度可能过高:将抗体进一步稀释至最佳浓度,并在4°C下进行孵育。
(2) 封闭不完全:延长封闭时间,并考虑更换封闭液。
(3) 洗涤不充分:增加各步骤洗涤时间与次数。
(4) 固定过程引起了自发荧光:如福尔马林/PFA通常会在绿色光谱中产生自发荧光,可尝试使用在红色光谱范围内发光的荧光团。
(5) 荧光显微镜参数设置不当:可通过调节荧光显微镜的参数来降低背景。
3)Q:荧光淬灭快?
A:荧光染色后应在1h内完成观察,放置4°C可保存4h,时间过长,可能会使荧光提前衰退。同时建议选择防荧光淬灭剂进行封片。
