泛素特异性蛋白酶(ubiquitin-specific proteases,USPs)是去泛素化酶(deubiquitylating enzyme,DUBs)家族最大的亚族,该族成员通过催化移除靶蛋白的泛素修饰,从而抑制靶蛋白的泛素-蛋白酶体途径降解或者影响靶蛋白的内吞、转运以及活性,是肿瘤干扰治疗的重要潜在靶标。
去泛素化蛋白酶(USP2),也称为UBP41和USP9,是一种细胞质蛋白,属于去泛素化酶肽酶C19家族。USP2广泛表达并作用于去泛素化选定的目标底物,涉及肿瘤、生物钟、代谢、炎症等过程。到目前为止,已知至少有三种USP2亚型,即USP2a(也称USP2-69),USP-41和USP2-45。其所有亚型都包含相同的催化核心,但在其N端和C端结构域内表现出变化。
USP2a是长度为605个氨基酸,预测分子量为68 kDa的规范亚型。人类USP2a与小鼠和大鼠直系同源物共享90%的氨基酸序列同一性。USP-41和USP-45的长度分别为362和396 个氨基酸,预测分子量为41和45 kDa。USP-41和USP2a已被证明是细胞凋亡和细胞增殖的关键调节因子。它们对多泛素化的靶蛋白(如FAS、MDM2和细胞周期蛋白D1)进行去泛素化,以促进或阻断细胞凋亡,具体取决于具体情况。USP-45的表达似乎受昼夜节律和营养线索的调节,被认为可调节肝糖异生和葡萄糖代谢。重组人去泛素化蛋白酶2氨基酸序列为第259到第605位的催化结构域。
