QS11在ARFGAP1酶实验和作为Wnt信号增效剂的EC50值分别为1.5 μM[1]和0.5 μM[2]。
嘌呤衍生物QS11是一种Wnt信号增强剂,抑制ADP核糖基化因子GTP酶激活蛋白1(ARFGAP1)。ARFGAP1促进ARF1结合的GTP水解,在膜运输和/或囊泡运输中起着关键作用。
体外:在HEK293细胞和人原代成纤维细胞中,QS11有效地增强Wnt活性(EC50值分别为0.5 μM和≥ 10 μM),而无显著细胞毒性。在含Wnt-3a的培养液中,2.5 μM QS11更能有效地激活Super(8X)TOPFlash报告基因(高约200倍)。当QS11与重组Wnt-3a蛋白(10 ~ 200 ng/ml)联合使用时,也可以观察到其协同作用[2]。
体内:在Wnt信号通路模型中,给予胚胎注射QS11、XWnt-8 RNA或QS11 + XWnt-8 RNA。相对于单一使用QS11和XWnt-8 RNA,XWnt-8 RNA和QS11联合使用显著地促进完整(20.9%)和部分(30.2%)双轴形成[2]。
临床试验:到目前为止,暂无可用的临床研究数据。
