Origami 2(DE3)感受态(Origami 2(DE3) Competent Cell)是一种可以高效表达外源蛋白的大肠杆菌感受态,本感受态可促进含有二硫键蛋白的正确折叠和增加蛋白的可溶性。
Origami 2(DE3)感受态的基因型为Δ(ara-leu)7697 ΔlacX74 ΔphoA PvuII phoR araD139 ahpC galE galK rpsL F′[lac+ lacIq pro] (DE3) gor522::Tn10 trxB (StrR, TetR)。本菌株是K12菌株的衍生菌株,其携带trxB和gor突变,这两个是还原途径的关键酶,其突变有利于二硫键的正确折叠,增加蛋白的可溶性。同时本菌株不含卡那霉素抗性,可用于卡那霉素抗性质粒的蛋白表达;本菌株具有链霉素和四环素抗性,为亮氨酸生长缺陷型。本菌株被λ噬菌体DE3溶原化,可同时表达T7 RNA聚合酶和大肠杆菌RNA聚合酶,因此可用于pET、pGEX和pMAL等载体的蛋白表达。
本产品提供即用型的Origami 2(DE3)化学感受态,同时提供pUC19质粒(2686bp,AmpR)作为模板,当使用pUC19质粒进行转化时,本感受态转化效率可达1 x 106 CFU/µg以上。本感受态经过特殊方法制备,最快可实现30 s快速转化,大大减少了操作时间,使用十分方便。(注:本产品采用特殊制备方法,肉眼观察为浅棕色,但不影响使用。)
