利用PRA测得,Nucleozin抑制MDCK细胞感染多种流感病毒,对A/WSN/33(H1N1),H3N2(临床分离株)和Vietnam/1194/04(H5N1)的EC50值分别为0.069±0.003 μM、0.16±0.01 μM和0.33±0.04 μM[1]。
流感病毒核蛋白在感染过程中表达最丰富,具有多种功能。Nucleozin有效抑制核蛋白于细胞核内积累,从而引起病毒死亡[1]。
体外:于MDCK细胞接种病毒后的6小时内,加入Nucleozin可以有效抑制病毒生长,这表明Nucleozin在病毒进入细胞与细胞核后的相关过程中发挥抗病毒作用。实际上,Nucleozin还影响多个核蛋白相关过程,尽管只有核蛋白进入到细胞核的过程比较明显[1]。然而,另一项体外研究表明,Nucleozin的靶点是病毒核糖核蛋白而不是核蛋白。此外,阻断病毒基因组在细胞质中的运输可以有效抑制IAV复制[2]。
体内:Nucleozin处理后的小鼠接种甲型流感病毒H5N1株(Vietnam/1194/04)后,与未经处理的对照组小鼠相比,具有相当高的存活率,显示出较强的体内抗病毒活性。所有未经处理的小鼠于接种后的第7天死亡。在Nucleozin治疗组中,给予Nucleozin(100 μl,2.3 mg/ml Nucleozin),每天两次,持续7天,50%的小鼠可以存活21天以上。动物研究结果表明,Nucleozin保护小鼠免受强毒性甲型H5N1流感病毒感染,有望用于抗流感病毒治疗[1]。
临床试验:到目前为止,Nucleozin仍处于临床前开发阶段。
