Lesinurad是URAT1的抑制剂,促进肾尿酸排出[1]。URAT1是一种调节尿酸从体内排出的肾脏转运蛋白[2]。
在临床研究中,Lesinurad有效抑制URAT1、OAT4以及另一种调节肾脏尿酸盐重吸收的转运蛋白。在一项研究中,21例高尿酸血症或痛风患者(sUA >= 8.0 mg/dL)随机分为Lesinurad组、别嘌呤醇非盲组或安慰剂组。大多数接受了Lesinurad治疗的患者于七天后出现sUA < 6 mg/dL,此效果与别嘌呤醇组患者出现的等同,但优于安慰剂组患者的效果。此外,Lesinurad在此项研究中显示出良好的耐受性[2]。与Probenecid(用于治疗肾功能不全)相比,Lesinurad可有效治疗轻度肾功能受损[1]。
