FK 3311是一种具有细胞透过性的可口服的磺酰,对环氧酶(COX)-2具有高选择性抑制,其IC50值为1.6 μM。
环氧酶(COX)-2是一种膜结合蛋白,将电子从细胞色素c转移到催化亚基1。COX-2的表达受多种介质的紧密调节和诱导,如细胞因子、生长因子和内毒素。
FK 3311选择性抑制人血小板和单核细胞中组成型和诱导型COX-2的活性[1]。
此化合物也广泛的用于各种动物模型中以研究COX-2的功能。在心肌缺血再灌注大鼠模型中,通过阴茎静脉给予FK 3311能显著降低血清中天冬氨酸转氨酶(AST)、乳酸脱氢酶(LDH)、丙氨酸转氨酶(ALT)和凝血恶烷(Tx) B2的水平。但是,FK 3311处理并没有降低大鼠体内6-酮-PG F1a水平[2]。另外,相较于对照组,FK 3311处理的大鼠肝组织血流量要显著改善。肝脏的组织学检验表明FK 3311处理能够降低肝组织损伤,并提高移植肝功能[3]。在犬肺移植模型中,FK 3311处理能够降低肺损伤、中性粒细胞肺浸润、锝-99m-白蛋白积累以及凝血恶烷B2的水平。FK 3311处理后的肺具有更好的肺部气体交换和血液动力学[4]。
