双荧光素酶报告基因系统是对基因调控表达进行研究的重要手段之一。本试剂盒中含有高纯度的萤火虫荧光素(Firefly Luciferin)和腔肠素(Coelenterazine),反应时可以发出对应波长的生物萤光 (bioluminescence),其中萤火虫荧光素在萤火虫荧光素酶、氧气、ATP和镁离子同时存在时发出波长为550-570nm的黄绿色光,而腔肠素在海肾荧光素酶和氧气存在时可反应发出波长480nm的蓝色光。
实验中先以荧光素为底物检测萤火虫荧光素酶(Firefly Luciferase)表达情况,后淬灭萤火虫萤光素酶的萤光信号,同时以腔肠素为底物检测海肾荧光素酶(Renilla Luciferase)的表达,实现双荧光素酶报告基因检测。
本品无需事先用裂解液将细胞裂解,再加入荧光检测试剂,简化了实验流程。使用时将Luciferase reagent直接加入细胞培养物中,使细胞裂解,并同时提供萤火虫荧光素酶底物,这使得本产品更加适用于高通量检测。我们的试剂适用于哺乳动物细胞培养基,比如含有1-10%血清的以下类型的培养基:RPMI 1640,DMEM,MEMα和F12等。
