白消安(busulfan)是一种DNA烷化剂[1].
DNA烷化剂连接于DNA的鸟嘌呤碱基,通过交联DNA双螺旋链的鸟嘌呤碱基,引起DNA损伤,从而抑制肿瘤的生长[1].
在正常的人二倍体WI38成纤维细胞中,白消安(busulfan)(7.5-120 μM)以剂量依赖的方式诱导衰老,这与长时间激活c-Jun氨基末端激酶(JNK)\p38丝裂原活化蛋白激酶(MAPK)和细胞外信号调节激酶(Erk)有关[2].衰老的诱导最开始是由细胞内的谷胱甘肽(GSH)瞬态损耗引起的,随后活性氧(ROS)生成增加,并激活 Erk和p38 MAPK通路[1].
在成年小鼠睾丸中,白消安(busulfan)诱导细胞凋亡并降低睾丸的重量.在第一周,细胞凋亡主要发生在精原细胞.在接下来的一周,精原细胞特异性标记Stra 8和c-kit减少,但Gli I保持不变,这表明分化的A型精原细胞凋亡[3].
