Bradford法的检测原理是在酸性介质中,考马斯亮蓝G-250与蛋白质的碱性和芳香族氨基酸特别是精氨酸结合,此时溶液转变为蓝色,溶液最大吸收峰从465 nm迁移到595 nm。依据溶液颜色的变化与蛋白质浓度成正比,可通过检测595 nm处的吸光度值对溶液中蛋白质浓度进行定量测定。
Bradford Protein Assay Kit是根据Bradford法研制而成,是蛋白定量检测常用方法之一,可实现快速、稳定和高灵敏度的蛋白浓度测定,并且检测数据有很好的线性关系。
本试剂盒有如下优点:检测速度极快,和BCA法相比,10-20个样品只需不足10分钟即可完成,大大缩短了检测时间;检测灵敏度高,可实现0.5 μg蛋白量检测;标准曲线线性关系好,5 μL体积的样品或标准品在0.1-1.5 mg/mL浓度范围内有良好的线性关系。
