BIBX 1382是一种用于癌症治疗的EGFR激酶抑制剂。使用在Sf9昆虫细胞中克隆和表达的HER2、VEGFR2、HGFR、IGF1R、B InsRK和c-src的胞质酪氨酸激酶结构域进行酪氨酸激酶活性测定,BIBX1382选择性地强效抑制EGFR激酶活性,IC50值为3 nM。使用EGFR高表达的A431细胞(vuval鳞状细胞癌)进行EGFR和pEGFR ELISA测定,BIBX1382抑制EGFR磷酸化,EC50为141 nM。[1] 在A431、FaDu(下咽鳞状细胞癌)和HN5(头颈部鳞状细胞癌)异种移植裸鼠模型中,10 mg/kg 日剂量的BIBX1382口服给药2周后显示强烈的剂量依赖性抗肿瘤活性。此外,50 mg/kg 日剂量的BIBX1382长期口服给药170天后,A431异种移植小鼠模型中肿瘤消退。[1]
