AM 1172是一种有效的稳定大麻素摄取的选择性抑制剂,IC50值为2.1 - 2.5 μM,也是脂肪酸酰胺水解酶(FAAH)抑制剂,Ki值为3.18 μM。
FAAH是丝氨酸水解酶家族的成员,是一种整合膜水解酶,可水解生物活性酰胺,包括将大麻素水解为游离脂肪酸和乙醇胺。在体外,FAAH具有酯酶和酰胺酶活性。在体内,该蛋白是用于分解称作脂肪酸酰胺(FAA)的生物活性脂类的主要分解酶。
在啮齿类大脑皮层神经元和人类星形细胞瘤细胞中,AM1172可阻断[3H]大麻素的内化,而非充当FAAH的抑制剂[1]。在小鼠大脑皮层神经细胞中,该物质也可阻断氚标记的AEA吸收,EC50值约为1.5 μM[1]。
关于AM 1172的体内效果,在人类或小鼠或其他动物模型中的研究仍待进行以提供相关证据。
