AC 45594是类固醇生成因子1(SF-1)的反相激动剂,IC50值为50-100 nM[1]。
SF-1是组成型的核激素受体,对性腺和肾上腺的发展至关重要,它充当性分化的整形因素。它主要是通过控制类固醇激素合成发挥作用[1]。
以阿霉素诱导的方式诱导人肾上腺皮质H295R/TR SF-1细胞过表达SF-1。AC 4559处理4天后,以剂量依赖性方式抑制基础细胞培养条件下的H295R/TR SF-1的增殖。10 μM的AC 45594显示出最大效应。SF-1的过表达诱导细胞增殖。在Dox存在条件下,经过1 μM的AC 45594培养后,显著降低了SF-1过表达的影响。此外,AC 45594增加sub-G1(凋亡)的细胞百分比[2]。在人肾上腺皮质细胞H295中,环磷酸腺苷能够诱导StAR的表达,这种诱导取决于SF-1的存在。AC 45594培养剂量依赖性地降低StAR mRNA的表达至基础水平。在蛋白质水平上,cAMP治疗24小时后,StAR增加了30倍。AC 45594治疗会剂量依赖性逆转cAMP依赖性的StAR的激活[1]。
没有发现任何关于AC 45594在动物体内作用的结果。
